In vitro culture technique of Camellia sinensis L to produce epicatehin with phosphor inducer
Abstract
In vitro culture techniques in several Asean countries are one of the cultivation technologies that can be applied in sustainable agriculture. Cultivation of in vitro culture of the Camellia sinensis L plant has been carried out for the purpose of obtaining the secondary metabolite of epicatehin which is also found in the Camellia sinensis L plant plucked from the field. The most difficult problems in in vitro culture technology included the acquisition of low epicatehin. To overcome the low epicatehin product problem, used method optimizing the phosphor inducer. The methods used were: (1) planting aseptically the slices of young leaf shoots of Camellia sinensis L plants on a culture-filled culture bottle, (2) re-cultivating the slices of young leaf shoots on the same medium with the addition of 2,4-D growth regulators, (3) optimizing the media with inducer  phosphor  by varying the concentration in the range: 40-200 ppm (4) evaluating in vitro culture through callus formation, (5) testing Camellia sinensis L callus to obtain qualitatively and quantitavely epicatehin secondary metabolites by high performance liquid chromatography methods. The result of the research in vitro culture of Camellia sinensis L could be production of epicatehin increases by 11.75 %
